PhysioLogics Tonalin® CLA provides multi-level diet support by working through distinct enzymatic and metabolic pathways. STRUCTURE AND FUNCTION
PhysioLogics Tonalin® CLA contains Conjugated Linoleic Acid (CLA), which exists as several isomers of the essential polyunsaturated fatty acid Linoleic Acid. Bioactive isomers include the cis-9, trans-11 isomer and trans-10, cis-12 isomer. Stereoisomers have seemingly different biological effects, making a precise mechanism of action difficult to discern. CLA has been shown to promote certain enzymes responsible for fatty acid betaoxidation and mobilization, such as hormone sensitive lipoprotein lipase. Preliminary evidence suggests that CLA promotes activation of peroxisome proliferatoractivated receptor alpha (PPARa), a regulator of lipid homeostasis. Other mechanisms discussed in the scientific literature include CLA's positive effects on total carnitine palmitoyl-transferase activity, promoting fatty acyl CoA transfer from the cytosol into the mitochondrial matrix. Uncoupling protein gene expression, as evidenced in murine models, may also be partially responsible for the metabolic effects of CLA at the level of ATP formation.


CLINICAL EVIDENCE
In a double-blinded trial, 60 male and female subjects who met the inclusion criteria of BMI>25 were randomized to receive one of four doses of Tonalin® CLA or matching placebo. Statistical analysis using Dunnett's test to make pair-wise comparisons of CLA vs. placebo revealed significant differences in body composition even at the lowest dose of 1.7g CLA per day. (Blankson et al, 2000) A double-blinded trial randomized 20 males and females to receive placebo or 1.8g per day of CLA from Tonalin®. Body composition was found to be significantly improved at 4, 8, and 12 week measures within CLA subjects and as compared to placebo subjects. (Thom et al, 2001)
